    Product Description

The Bisulfite-Converted Universal Methylated Human DNA Standard is designed to be used as a control for DNA bisulfite conversion and downstream analyses including PCR, MSP, and other amplification-based assays. This DNA is identical to our Universal Methylated Human DNA Standard, but has been bisulfite-converted using the EZ DNA Methylation – Direct (D5020) kit. This product is provided as a 20 ng/µl solution and contains enough for 50 reactions. The primer set included with the standard has been designed and validated to amplify a segment of the bisulfite-converted DNA.

    Resources

The sample is derived from human male brain tissue.

All standards are enzymatically methylated by CpG Methylase in vitro at all CpG dinucleotides. The methylated DNA is very highly methylated (> 95% of all CpG sites).

D5015 is fully methylated human DNA which has been bisulfite converted using the EZ DNA Methylation-Direct Kit. When run in parallel with experimental samples through library prep, PCR, etc., this standard can be used to determine if there are issues with sample quality or conversion efficiency that may be negatively impacting downstream applications.

This standard is included with primers targeting human MLH1 mismatch repair gene, and will amplify non-methylated, methylated, and mixed methylated templates. By amplifying bisulfite converted DNA with the included primers, researchers can test and optimize their bisulfite PCR conditions.

We recommend ZymoTaq DNA Polymerase, a hot start Taq polymerase specifically designed to amplify bisulfite converted DNA. Another hot start DNA polymerase of choice may also be used.

The standards are stable at ≤ -20°C for up to 24 months. For long term storage standards should be stored at ≤ -70°C.
